Samuel Amo-Ameyaw played for Tottenham Hotspur during his youth career.

He was selected on Southampton's subs bench for the first time on 28 May 2023 in a Premier League game against Liverpool. Aged 16 years and 314 days, he came on as a late substitute to become the youngest-ever player to appear in the Premier League for the Saints; previously Luke Shaw at West Bromwich Albion on 5 November 2012 (17 years and 116 days).
